Pipette Calibration is vital to any laboratory. A pipette functions to transfer or dilute precise volumes of fluid. Pipette calibration ensures pipettes are working to defined regimes or work protocols that meet or exceed manufacture specifications.

As good practice an operator should always be trained in the correct use of a pipette and occasionally compare himself against standards to ensure good practice. Note that temperature and humidity can affect a pipettes performance.

Micro Quality Calibration uses the gravimetric analysis which entails dispensing samples of distilled water into a receiving vessel in a precision analytical balance. The density of water is a known constant, the temperature, barometric pressure and humidity are recorded (the Z-factor used in the final mass calculation) and kept within certain limits and thus the mass of the dispensed sample provides an accurate indication of the volume dispensed.

There are 2 basic types of calibration, as found where the pipette is just tested in the "as found" condition with no adjustment and the "as calibrated" where the pipette is cleaned, serviced, calibrated and the results given in the report. The 2 types may be combined into a as found/ as calibrated report. MQC's ISO 17025 scope of accreditation covers the repair and calibration of pipettes. There are 3 important measurement's:

·         Mean error or accuracy is the amount the mean of the samples varies from the nominal volume selected usually expressed in a percentage

·         Repeatability or precision is an assessment of the standard deviation or coefficient of variation between the highest and the lowest sample

·         Uncertainty can be stated that no measurement is 100% correct. There are factors that contribute to a measurement’s uncertainty, or ‘doubt’. To be clear, this is not error. Error is the span of a measurement from the target setting and is correctable.
